Abstract

The adulteration of plants and their materials used in herbal formulations poses a severe health concern. There is a need for a reliable, cost-effective, and robust molecular biomarker to distinguish among plant species and identify herbal plants and raw drugs from adulterants.

Aim of the study

The study aims to develop a strategy for identifying novel DNA-based markers using suppressive subtractive hybridization and next-generation sequencing to distinguish between authentic medicinal plants and their adulterants.

Methodology

  • Taxonomic identification and herbarium preparation was performed for B. diffusa, T. portulacastrum, T. cordifolia, and T. sinensis.
  • Molecular authentication using universal markers (nrITS and matK) through Sanger DNA sequencing
  • Suppressive subtractive hybridization and Next-generation sequencing technology were used to develop DNA markers.
  • Validation was performed using Sanger sequencing.

Results

  • Suppressive subtractive hybridization and next-generation sequencing were used to generate species-specific markers for B. diffusa and T. cordifolia and tested them in different accessions of these plants. Four primers specific to B. diffusa and one specific primer for T. cordifolia were obtained.
  • The developed primers showed 100% sensitivity and specificity for the tested accessions of B. diffusa

Conclusion

The DNA markers developed using suppressive subtractive hybridization and next-generation sequencing technology can authenticate Boerhavia diffusa and Tinospora cordifolia plants and distinguish them from their adulterants with 100% sensitivity and specificity. These DNA markers can identify and differentiate these plant species in herbal formulations, addressing the issue of adulteration in herbal medicines.
